GPP: generalized pustular psoriasis IL-36RN: interleukin-36 receptor antagonist PV: psoriasis vulgaris INTRODUCTION Generalized pustular psoriasis (GPP) is a rare but serious inflammatory skin disease that may be caused by deficiency of interleukin-36 receptor antagonist in which there is presence of homozygous or compound heterozygous mutations of the IL-36RN gene. Most GPP cases are sporadic, although some may be autosomal recessive. A significant proportion of GPP without psoriasis vulgaris (PV) had homozygous or compound heterozygous mutations in IL-36RN. GPP alone is a distinct subtype and is etiologically distinguished from GPP with PV. Most GPP alone is caused by deficiency of the IL-36RN caused by IL-36RN mutations, in contrast to GPP associated with PV, in which this mutation is usually absent.

Introduction Homozygous missense mutation in the IL36RN gene resulting in deficiency of interleukin-36-receptor antagonist (DITRA) is phenotypically presented as severe generalized pustular psoriasis starting in early childhood. Compound heterozygous cases have been described with the same DITRA phenotype, but to our knowledge heterozygous IL36RN mutation related to severe generalized pustular ...

Introduction Interleukin-36-receptor antagonist deficiency (DITRA) is a recently described auto-inflammatory disease, characterized by repeated flares of generalized pustular psoriasis, high fever, asthenia and systemic inflammation. This condition is caused by homozygous missense mutation in the IL36RN gene, encoding the interleukin-36-receptor antagonist (IL-36Ra), an anti-inflammatory cytoki...

YM is the first son of Tunisian consanguineous parents who developed, at 2 weeks of life, an erythematous and scaly eruption, with subsequent rapid evolution toward generalized pustular psoriasis. Afterward, cutaneous flares of diffuse erythematous rash and pustules involving the whole body appeared, with a once weekly periodicity. Intense irritability was present during flares without fever. M...

Generalized pustular psoriasis is a severe skin disease characterized by epidermal hyperplasia, neutrophil-rich abscesses within the epidermis, and a mixed inflammatory infiltrate in the dermis. The disease may be caused by missense mutations in the IL-36 receptor antagonist, IL-36Ra. Curiously, the related IL-1Ra has therapeutic effects in some of these latter patients. Here, using an experime...
